DESCRIPTION
Western blotting is an analytical technique to detect the levels of expression of desired proteins in a given sample. The proteins of interest are run on an SDS PAGE gel and then the proteins are transferred from the gel to a membrane with the help of electroblotting. In this technique, the proteins contained in the gel are sandwiched between membranes (such as PVDF of Nitrocellulose). An electric field is applied and the negatively charged proteins migrate to the positive electrode and in the process, get transferred onto the membrane, where it becomes available for further analysis. In the case of semi-dry blotter, the membranes are placed in close contact with the electrodes to ensure faster and more efficient transfer of bands. The system required minimal buffer. It is only required to equilibrate the membranes and filter papers with the transfer buffer before the blotting begins. In such systems, it is essential to remove any unwanted moisture.
