Description
Proteinase K, supplied as lyophilized powder, is a broad spectrum serine protease useful for general digestion of proteins. This is one of the most active endopeptidases known that does not exhibit any pronounced cleavage specificity. The enzyme is effective on native proteins and can be used to rapidly inactivate RNases and DNases during nucleic acid isolation, sequencing, PCR and RT-PCR. The enzyme is active even in presence of SDS, EDTA or urea and has a pH – optimum of 7.5 to 10.5. Complete activity is maintained over several hours in pH range 6.5 to 9.5. Proteinase K is inhibited by PMSF.
Unit definition:
One unit of Proteinase K produces 1 mmole of Folin-positive amino acid(s) in 1 min at 37°C.
Activity: 33.79 U/mg Protein.
Directions to use: Prepare 20 mg/ml stock solution in deionized water and use at concentration of 50 µg/ml at 50°C to 60°C.
Store: -20°C
Note: To avoid frequent freeze-thaw cycles, store stock solution as aliquots at -20°C
